Calcium can also be released genetic code into the cell from the sarcoplasmic reticulum (SR) by inositol 1,4,5-triphosphate (IP3) and Ca2 + -stimulated calcium genetic code release The processes genetic code on the left side increase cytoplasmic calcium and promote contraction;. Those on the . right side decrease internal genetic code calcium and promote relaxation DAG, diacylglycerol; IP3, inositol 1,4,5-trisphosphate; PIP2, phosphatidylinositol 4,5-bisphosphate.

Introduction According to statistics, in all causes of infertility among the causes of male infertility about the proportion dna of about 45%, and female infertility reasons dna for the same. The so-called male infertility refers to the lack of their sperm count, poor sperm motility, sperm abnormal appearance, dna or will itself generate sperm manufacturing problems, infertility caused. For people with severe male infertility couples, even if IVF is often because they can not let the wife's egg is fertilized, the embryo can be implanted faced no dilemma, ultimately, to do IVF using donor sperm or adoption of another child . Thanks to advances in artificial reproductive technology, the egg cytoplasm single sperm microinjection technique (Intracytoplasmic Sperm Injection referred ICSI), has been widely used in the treatment of male infertility, get a good pregnancy rate and implantation rate, become men Infertility savior.
Professor in the egg cytoplasm single sperm microinjection technique (ICSI) 1992 Belgium Free University Van Steirteghem take a little sperm of male infertility patients, the use of micro-injection technology within dna a single sperm injected into the egg cytoplasm, successfully fertilize an egg, and pregnancy gave birth to the baby. The egg cytoplasm single sperm microinjection (Intracytoplasmic Sperm Injection referred ICSI) technique allows the treatment of male infertility has entered a new milestone. Male infertility can be found as long as there is a sperm survival, the use of ICSI technique it is possible to let the egg is fertilized and then pregnancy.
The whole process is done in a micromanipulator micromanipulator (Micromanipulator) on (Figure 1). While using a fixed micro tubules (Holding pipette), using a negative pressure to suck the eggs, sperm and then applied on the other side of the micro-injection of a small tube (Injection pipette), the syringe is injected into the egg in the microstructure in order to achieve the purpose of fertilization. Before sperm microinjection, sperm must first be carried dna out without automation (Immobilized), dna usually applied injection fine tubules, gently dna draw at the tail of the sperm, let not move, then sperm injection inhalation of fine tubules, then apply a fixed fine tubules withstand the egg, then sperm injection containing fine within a small tube, the egg polar body (Polar dna body) adjustment at 6 o'clock or 12 o'clock dna position, then align the egg 3 o'clock position, the sperm injected into the egg inside, complete ICSI procedure (Figure 2). With polarizing optical system (Polarizing dna Optical System) to operate ICSI, can reduce dna the possible damage during the operation a small tube of egg injection of fine quality skeleton.
Applies mainly targeted male factor infertility couples include: (a) azoospermia (azoospermia). (2) sperm rare disease (Oligospermia). (3) poor sperm fertilization, including dna sperm unable to disease, abnormal sperm, poor motility, sperm zona pellucida binding obstacles. (4) congenital testicular sperm manufacture bad --- can be obtained by testicular dna sperm sperm. (5) congenital defects --- vas deferens sperm can be obtained via the epididymal sperm. (6) has the patient anti-sperm antibodies. (7) cases have done IVF IVF failure. (8) Part of unexplained infertility patients
When ICSI ICSI technique is safe just come out, because with artificial sperm directly into the egg cytoplasm, dna the process than natural fertilization process more aggressive, so it is worrying security. In the natural fertilization dna process, allowing the sperm to fertilize the egg stand out from tens of millions of sperm in the elite only, but ICSI is a technician in the laboratory by a randomly selected sperm injected into the egg cytoplasm to a clear violation natural principles. However, Professor Van Steirteghem track 201 ICSI baby to two years of age, and with them the other 131 to compare traditional IVF physical and intellectual development, found that two dolls and no significant differences. To further understand the safety of ICSI, the European Infertility Medical Association integration 24 countries a total of 90 infertility centers, tracking 807 ICSI babies, including 16 babies major congenital malformations, the probability of occurrence is 1.9%, with natural conception and birth of the baby did not increase compared to the probability of congenital malformations. However, dna due to too many variables statistically, currently dna unable to make any conclusions. Of course, these studies will continue to be tracked down, because dna of these reproductive disadvantaged groups to use ICSI procreation, yes right? Is wrong? There is still some controversy, the future need to clarify further.

Pregnancy and delivery of a normal infant following transfer of a single preembryo from an ICSI cyc

Fertil Steril multaq 2004 Dec; 82 (6):.. 1662-5 Normal birth after single-embryo transfer in a patient with excessive polypronuclear zygote formation multaq following multaq in vitro fertilization and intracytoplasmic sperm injection.
Pregnancy and delivery of a normal infant following transfer of a single preembryo from an ICSI cycle in which only one zygote multaq showed a normal pronuclear number (2PN) and 12 zygotes appeared polypronucleated (> or = 3PN) RESULT (S).:
On the first IVF cycle, 23 oocytes were retrieved and inseminated with 240 x 10 (3) motile multaq sperm / mL, after which two zygotes showed a normal pronuclear number and 20 zygotes appeared polyploid with three to seven pronuclei. Transfer of two poor-quality day-3 preembryos following assisted hatching multaq did not achieve pregnancy. On the subsequent ICSI cycle, 33 oocytes were retrieved, and 17 mature oocytes were subjected to ICSI, after which only one zygote showed 2PN and 12 zygotes appeared polyploid with three to eight pronuclei. The normally fertilized zygote developed into a poor-quality, day-3 embryo and was subjected to assisted hatching Transfer of this preembryo resulted in an uneventful pregnancy and birth of a normal infant multaq CONCLUSION (S)..:

The objective of this research was to investigate the effects of cooling on the development of bovine zygotes. One-cell bovine embryos were maintained at 39 degrees C (control), 20 degrees C, 10 degrees C, or 0 degree C for 5, 10 , or 20 minutes, then cultured in vitro for 7 days and the proportion of embryos developing to the compact morula or blastocyst stage compared between different treatments. Duration of exposure time had no effect on development. Development rates to the compact morula or blastocyst stage were 3.9%, 11.4%, 17.4%, and 24.4% for zygotes maintained at 0 degree C, 10 degrees C, 20 degrees C, and 39 degrees C, respectively, with differences in embryo yield between every treatment biogen (P <0.05). In a second experiment, bovine pronuclei (karyoplasts) and cytoplasts were cooled at 0 degree C or maintained biogen at 39 degrees C for 5 minutes Pronuclear transplantation was then utilized to create 4 types of reconstructed embryos, those with:. 1) non-cooled pronuclei biogen and non-cooled cytoplasm, 2) non-cooled biogen pronuclei and cooled cytoplasm, 3) cooled pronuclei and non-cooled cytoplasm, and 4) cooled pronuclei and cooled cytoplasm. The proportion of embryos developing to the blastocyst biogen stage was highest when non-cooled pronuclei were transferred into non-cooled cytoplasm (18.9%), and similar to that of non-cooled, non-manipulated control zygotes (13.2%, P> 0.05). No embryos developed to the blastocyst stage when pronuclei (cooled or non-cooled ) were transferred into cooled cytoplasm. However, zygotes with cooled pronuclei transferred into non-cooled cytoplasm yielded 4.5% blastocysts (P <0.05). More embryos developed to the compact morula or blastocyst stage when non-cooled vs. cooled cytoplasm was utilized, regardless of whether the pronuclei were cooled (P <0.05). These data demonstrate that pronuclei are more tolerant to low temperature exposure than is ovum cytoplasm.

1 calcium pump (calcium pump) need to consume energy drosophila melanogaster recovery Ca2 +, Ca2 + recycling to the sarcoplasmic reticulum or outside the cell. In different cell recovery mechanisms of Ca2 + is not the same, is divided into the following three: A. plasma-membrane Ca2 + -ATPase (PMCA): in the cell membrane calcium pump (calcium pump), cardiac muscle, smooth muscle this mechanism Lord B. secondary active transport: the use of Na + concentration gradient into the intracellular Ca2 + will send out C. Sarcoplasmic and endoplasmic reticulum calcium ATPase (SERCAs): In the endoplasmic reticulum or sarcoplasmic reticulum calcium pump (calcium pump), skeletal muscle this mechanism based 2. Ca2 + binding calcium and protein needed to type non-covalent binding to the cells, so reducing the number of free Ca2 +, helping calcium pump (calcium pump) is easy to Ca2 + send them. (2) GSK-3 (kinase A, glycogen synthase kinase-3): will be phosphorylated (phosphorylate), the degradation of the β -catenin itself (degradation), that is to say, GSK-3 is inhibited β -catenin alexion performance, it is also with β -catenin alexion exist in the cytoplasm of the egg were. (3) GSK-3 inhibitor exists only in the cytoplasm of the vegetal pole cortex (vegetal cortex), and after the eggs are fertilized, GSK-3 inhibitor will move along microtubules, moved to the gray crescent and prevent the degradation of β -catenin (through GSK-3 reduces the concentration). This mechanism will result in relatively high concentrations of β -catenin in the dorsal, play an important role in the signaling system between cells, after which you can start a series of reactions on cell selection and embryonic development. I have commented out with # two lines iptables -I INPUT 1 -p udp --dport 1194 -j ACCEPT iptables -I

There are various circumstances, access to the home within the LAN from outside the home was needed. I tried to set where it is available (Linksys WRT150N) wireless LAN router where you installed the firmware that made the VPN build of DD-WRT. Web site you reference is a thing of the next. Important information thank you. To the remote access router -OpenVPN to try playing VPN (the easy way) in v24 + (English) http://www.dd-wrt.com/wiki/index.php/VPN_%28the_easy_way%29_v24%2B DD-WRT how to create a certificate (Japanese) http://linuxandxx.blog.fc2.com/blog-entry-47.html server protean was able to create without problems by the method of reference web site mentioned above. There are two access methods types of OpenVPN. Bridge system and (Routing) routing scheme is (Bridging), but there was a description set is the easy routing scheme. But I was not able to be installed easily in relation to the location of the home LAN of the OpenVPN server. They will work fine if the installation protean method The wireless LAN router that was installed OpenVPN server is like making a connection to the external Internet. [Wireless LAN router] (OpenVPN server) [home server class] towards the bridge system was the first successful in that (web server, mail server) surprising protean [Internet] arrangement to work with routing scheme. There was no problem in Reference web site as per above this. It was only configured to use the tap instead of tun as a device in the client configuration of the machine from which you want to access from outside protean As a side note. Well The placement of the home LAN at home. Wireless LAN router you have installed the DD-WRT uses as a wireless LAN access point as (disable) disable the WAN terminal. Placement of my home [Internet] domain name: example.com [NTT West Hikari HGW] (PR-400NE) (192.168.24.1) protean over over over over over over over over over over over over over over over [wireless protean LAN router] (OpenVPN) [home server] (192.168.24.8) (192.168.24.100) - home server management DNS, DHCP, WEB, MAIL, the Asterisk - routing scheme of the arrangement of our house This is the setting of the OpenVPN DD-WRT on. Here was also done in reference to the reference web site mentioned above, but I have some change.
I have revised the definition of the items in the push settings [OpenVPN Config] - [OpenVPN Daemon]. The home server directed to (192.168.24.100) the setting of the DNS server was so as to be registered to the client machine domain name (example.com) further. It had been registered automatically to the resolv.conf on the client machine in this push setting.
push "route 192.168.24.0 255.255.255.0" push "dhcp-option DNS 192.168.24.100" push "dhcp-option DOMAIN example.com" server 192.168.66.0 255.255.255.0 dev tun0 proto udp keepalive 10 120 dh / tmp / openvpn / dh.pem ca /tmp/openvpn/ca.crt cert /tmp/openvpn/cert.pem key /tmp/openvpn/key.pem # Only use crl-verify if you are using the revoke list - otherwise leave it commented out # setting screen of 5001 OpenVPN crl-verify /tmp/openvpn/ca.crl protean # management parameter allows DD-WRT's protean OpenVPN Status web page to access the server's management port # port must be 5001 for scripts embedded in firmware to work management localhost
I have commented out with # two lines iptables -I INPUT 1 -p udp --dport 1194 -j ACCEPT iptables -I FORWARD 1 -s 192.168.66.0/24 -j ACCEPT Firewall set a bottom. After this, OpenVPN packets from the external Internet: Perform protean the [IPv4 packet filter settings] and [static IP masquerade Settings to be directed to (192.168.24.8) wireless LAN router (UDP 1194) has become a OpenVPN server You. It has become that it is connected properly in the status screen of OpenVPN. I tried doing actively reviewing the routing configuration of the DD-WRT wireless protean LAN router protean at first, but was not able to access. Look for the case "has installed protean OpenVPN server in your LAN" on the net, I tried to search the information that would be helpful or what. Then, that there is a problem with the routing configuration protean of the router HGW home LAN of (PR-400NE) Surprisingly was found. I did not must be set in the routing HGW (PR-400NE) a packet of 192.168.66.0/24 is a virtual LAN of OpenVPN. Was a surprising blind spot (sweat). As to route to (192.168.24.8) wireless LAN router is the gateway of OpenVPN OpenVPN protean packet of the (192.168.66.0/24) [static routing configuration - the configuration screen of HGW (PR-400NE) [Advanced] I was set to. For more information, please refer to the image of the setting screen. [Advanced Settings] - [static routing configuration] [Advanced Settings] - [Static Routing Settings] - [Entry Edit] it can be plugged back in from the client machine again in this state, to access the home server properly now.
